چکیده
Martin Matzuk, director of the Center for Drug Discovery at Baylor College of Medicine, recalls a childhood visit to the New Jersey office of orthopedic surgeon Joseph Lepree. “I was always impressed at how full his waiting room was and the time that he spent examining me and speaking with my father,” says Matzuk. “On one visit, he askedmewhat I wanted to bewhen I grew up, and I said, ‘a doctor, just like you.’ From that point on, I knew I was going to be a physician.” Today, Matzuk is a clinical pathologist and endowed chair in Baylor’s department of pathology and immunology. For nearly three decades, Matzuk’s research has focused on the critical proteins and mechanisms involved in reproductive development, leading to advancements in the treatment of infertility and reproductive cancers, such as ovarian cancer. Matzuk and his team have also begun to characterize small-molecule contraceptives targeting the male germ line, paving the way toward the creation of the first effective and nonhormonal birth control pill for men. In recognition of these and other achievements, Matzuk was elected to the National Academy of Sciences in 2014.

Appetite for reproduction: dietary restriction, aging and the mammalian gonad
The major physiologic theory of aging, the disposable soma theory, links dietary restriction (DR), also known as calorie or food restriction, to prolonged lifespan and makes specific predictions about the effects of aging and DR on reproduction. A recent study in BMC Biology profiling the effects of aging and DR on gonadal gene expression provides novel molecular evidence that has a significant...
متن کاملDeconstructing mammalian reproduction: using knockouts to define fertility pathways.
Reproduction is the sine qua non for the propagation of species and continuation of life. It is a complex biological process that is regulated by multiple factors during the reproductive life of an organism. Over the past decade, the molecular mechanisms regulating reproduction in mammals have been rapidly unraveled by the study of a vast number of mouse gene knockouts with impaired fertility. ...

Lipid storage diseases are debilitating inherited metabolic disorders that stem from the absence of specific lysosomal enzymes that degrade selected lipids. Most characteristically, these disorders affect the nervous and the reticulo-endothelial systems, with massive organomegaly resulting from the presence of engorged, lipid-laden macrophages. In this issue of the JCI, Yildiz et al. describe t...
متن کاملNOBOX deficiency disrupts early folliculogenesis and oocyte-specific gene expression.
Primordial ovarian follicles in mice form when somatic cells surround individual oocytes. We show that lack of Nobox, an oocyte-specific homeobox gene, accelerates postnatal oocyte loss and abolishes the transition from primordial to growing follicles in mice. Follicles are replaced by fibrous tissue in female mice lacking Nobox in a manner similar to nonsyndromic ovarian failure in women. Gene...
